摘要

肿瘤已成为全球范围内威胁人类健康的主要公共卫生挑战之一。黄烷醇作为一类广泛存在于茶叶、可可、水果等日常膳食中的多酚化合物,因其潜在的抗肿瘤活性备受关注。细胞与动物实验进一步揭示,黄烷醇可通过抑制细胞增殖、诱导凋亡、阻滞细胞周期、抑制肿瘤侵袭与转移、调节氧化应激与免疫应答等发挥多靶点、多途径的抗肿瘤分子机制。大量流行病学研究表明,膳食黄烷醇摄入与多种肿瘤风险降低相关。其特殊的构效关系,特别是聚合度和没食子酸酯化对其抗肿瘤活性具有关键的影响。然而,由于黄烷醇具有溶解度较低、化学稳定性较差以及口服生物利用度极低的特点,其应用尚面临巨大的挑战。为突破此瓶颈,纳米递送系统等新兴策略通过改善药物溶解性、提高稳定性、增强肠道吸收与肿瘤靶向性,展现出提升黄烷醇生物效价的巨大潜力。本文系统综述了黄烷醇在肿瘤防治领域的研究进展,重点阐述其多机制的抗肿瘤作用,同时展望了纳米技术等新型递送策略在提升其生物效价和临床转化潜力中的应用前景,为黄烷醇的进一步应用提供全面的理论依据。

Abstract

Tumors have become one of the major public health challenges threatening human health worldwide. Flavanols, as a class of polyphenolic compounds widely found in daily diets such as tea, cocoa, and fruits, have attracted attention for their potential antitumor activities. Cellular and animal studies further reveal that flavanols exert multi-target and multi-pathway antitumor molecular mechanisms, including inhibiting cell proliferation, inducing apoptosis, arresting the cell cycle, suppressing tumor invasion and metastasis, and modulating oxidative stress and immune responses. Numerous epidemiological studies indicate an association between dietary flavanol intake and a reduced risk of various cancers. Their specific structure-activity relationships, especially the degree of polymerisation and gallic acid esterification, have a critical impact on their anti-tumor activities. However, the application of flavanols is still facing great challenges due to their low solubility, poor chemical stability and very low oral bioavailability. To overcome these bottlenecks, novel strategies such as nanodelivery systems have shown great potential in enhancing the bioefficacy of flavanols by improving drug solubility, increasing stability, enhancing intestinal absorption, and achieving tumor targeting. In this paper, we systematically review the research progress of flavanols in the field of tumor prevention and treatment, with a focus on elucidating their multi-mechanistic antitumor effects. And we look forward to the prospects of novel delivery strategies such as nanotechnology in enhancing their bioefficacy and clinical translational potentials, to provide a comprehensive theoretical basis for the further application of flavanols.
